Overview

The B-Raf family includes ARAF, BRAF, and CRAF (RAF1), which are serine/threonine kinases that play a central role in the RAS-RAF-MEK-ERK signaling pathway. This pathway regulates cell growth, differentiation, survival, and development. Dysregulation, particularly mutations in BRAF, is a major contributor to human cancer. Germline mutations in BRAF or CRAF can cause developmental syndromes known as "RASopathies".
