Polyclonal tumor antigens from B16F10 cells represent the collective set of immunogenic molecules, including tumor-associated antigens (TAAs) and mutation-derived neoantigens, found in the B16F10 murine melanoma cell line (Overwijk & Restifo, 2001). This cell line is a standard model for studying tumor immunology and developing cancer vaccines due to its poor immunogenicity and high metastatic potential. Key specific antigens within this polyclonal mixture include gp100 (Pmel), tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2/Dct), which are involved in melanin synthesis (Bloom et al., 1997). Additionally, the mixture contains unique neoantigens resulting from somatic mutations specific to the B16F10 line (Castle et al., 2012). Therapeutic strategies involve using these antigens to train the immune system to recognize and destroy melanoma cells, often delivered via lysates, viral vectors, or mRNA platforms. By utilizing a polyclonal approach, researchers aim to overcome tumor heterogeneity and prevent immune escape by inducing a broad T-cell response against multiple targets simultaneously (Schreurs et al., 2000). This multi-target strategy is particularly relevant for addressing the challenges of immune tolerance and the low immunogenicity of individual self-antigens.
