Overview

B7-related protein 1 (B7RP-1), also known as ICOS Ligand (ICOS-L) or CD275, is a type I transmembrane glycoprotein belonging to the B7 family of immune-regulatory ligands and immunoglobulin superfamily. It primarily expressed on antigen-presenting cells (APCs) such as dendritic cells, macrophages/monocytes, and some activated B cells, where it interacts with its receptor, ICOS, on activated T cells. This interaction provides a crucial co-stimulatory signal for T cell activation, differentiation (especially Tfh cells), germinal center formation, and antibody production. As such, B7RP-1 is an area of focus in immunotherapy, with potential applications in enhancing anti-tumor immunity and vaccine efficacy. However, dysregulation may contribute to autoimmune diseases by causing excessive immune responses. Therefore, therapeutic targeting requires careful consideration of potential safety concerns related to immune-related adverse events and exacerbation of autoimmunity.
