Overview

BAALC antisense RNA 1 (BAALC-AS1) is a long non-coding RNA (lncRNA) located in the antisense strand of the BAALC gene region on chromosome 8q22.3[2]. It is also known as lncFZD6, FZD6-DT, FZD6 divergent transcript, or LOC105369147. BAALC-AS1 is upregulated in esophageal squamous cell carcinoma (ESCC) and plays an oncogenic role by promoting cell proliferation, migration, and invasion. Mechanistically, BAALC-AS1 directly binds to the protein G3BP2 and inhibits G3BP2-dependent degradation of c-Myc mRNA, leading to increased c-Myc expression. In turn, c-Myc upregulates BAALC-AS1 transcription, establishing a positive feedback loop. High BAALC-AS1 expression correlates with aggressive tumor phenotypes and is proposed as a potential therapeutic target and a biomarker for cancer diagnosis and prognosis, with particular emphasis in ESCC[1][2][3][4]. There are currently no approved drugs targeting BAALC-AS1.

Mechanism of action

No known approved drugs directly targeting BAALC-AS1. Experimental evidence: silencing BAALC-AS1 suppresses c-Myc signaling, reduces proliferation and invasion in ESCC cells[1][2][3][4]

03

Biological functions

Regulation of gene expressionPromotion of cell proliferationPromotion of cell migrationPromotion of cell invasionPositive regulation of c-Myc expression
04

Disease associations

CancerEsophageal squamous cell carcinoma (ESCC)Potential role in other malignancies (pending further validation)
05

Safety considerations

No established safety concerns, as clinical targeting is preclinical; potential safety issues would relate to general targeting of non-coding RNAs and interfering with c-Myc regulation
06

Biomarkers

Overexpression of BAALC-AS1 as a biomarker for ESCC malignancy and prognosis[1][2][3][4]
