Overview

The bacterial 30S ribosomal subunit decoding center is a critical functional region within the small ribosomal subunit that ensures the accuracy of protein synthesis. It is primarily composed of the 16S ribosomal RNA (rRNA), specifically the aminoacyl-tRNA binding site (A-site), where it monitors the fidelity of the codon-anticodon interaction (Ogle et al., 2001, Science). During translation, conserved nucleotides A1492 and A1493 flip out to sense the minor groove of the mRNA-tRNA helix, triggering the conformational changes necessary for peptide bond formation (Vicens & Westhof, 2003, ChemBioChem). This site is the primary target for aminoglycoside antibiotics, which bind to the 16S rRNA and stabilize the "on" state of the decoding center, leading to the incorporation of incorrect amino acids and subsequent bacterial cell death (Kohanski et al., 2010, Nature Reviews Microbiology). Other antibiotics, such as tetracyclines, target this region by sterically blocking the entry of aminoacyl-tRNA into the A-site (StatPearls, 2023). While highly effective, drugs targeting this center can cause significant side effects like ototoxicity and nephrotoxicity due to structural similarities between bacterial 16S rRNA and human mitochondrial 12S rRNA (Hobbie et al., 2008, PNAS).
