The bacterial 30S ribosomal subunit decoding center A-site is a critical functional region within the 16S ribosomal RNA (rRNA) that ensures the accuracy of genetic translation [1, 7]. Located at the interface of the 30S subunit, this site monitors the interaction between the mRNA codon and the incoming aminoacyl-tRNA anticodon [4, 16]. During correct decoding, conserved nucleotides A1492 and A1493 flip out of helix 44 to stabilize the codon-anticodon helix, a process essential for high-fidelity protein synthesis [7, 15]. This site is the primary target for aminoglycoside antibiotics, which bind to the internal loop of the A-site and lock the decoding nucleotides in the "flipped-out" state [1}
