The bacterial biofilm extracellular polymeric substance (EPS) matrix and microbial cell membranes constitute a synergistic structural complex that protects bacterial communities from external threats. The EPS matrix is a self-produced mixture of polysaccharides, proteins, lipids, and extracellular DNA (eDNA) that facilitates surface attachment and acts as a physical barrier against host immune cells and antibiotic penetration (Flemming & Wingender, 2010, Nature Reviews Microbiology). Within this matrix, the microbial cell membranes serve as the essential metabolic boundary, maintaining ion gradients and providing the site of action for many membrane-active antimicrobials. This dual-layered defense system is a primary driver of chronic and persistent infections, as it allows bacteria to survive in a dormant, highly tolerant state (Costerton et al., 1999, Science). Therapeutic interventions targeting these structures aim to either enzymatically dissolve the EPS 'glue' to release bacteria into a planktonic state or directly disrupt the membrane integrity of the encased cells to induce lysis (Donlan, 2002, Emerging Infectious Diseases). Understanding the interplay between the matrix and the membrane is critical for developing strategies to combat multidrug-resistant biofilm-associated diseases.
