Overview

The bacterial cell envelope is a complex, multi-layered structure that provides structural integrity and protects the bacterium from environmental stressors and osmotic pressure [1]. It typically comprises the inner cytoplasmic membrane, a peptidoglycan cell wall, and, in Gram-negative species, an outer membrane containing lipopolysaccharides [1][2]. This envelope is home to numerous essential enzymes, such as penicillin-binding proteins (PBPs), which are responsible for the synthesis and cross-linking of the peptidoglycan layer [2][3]. Because the components of the bacterial cell wall are unique to prokaryotes and absent in human cells, they represent ideal targets for selective toxicity [3]. Antibiotics like beta-lactams, glycopeptides, and polymyxins exert their effects by inhibiting cell wall assembly or disrupting membrane stability, ultimately leading to bacterial cell lysis and death [4]. Targeting the cell envelope remains a cornerstone of antibacterial therapy, though the rise of multi-drug resistant strains poses a significant challenge to current treatment regimens [5].
