The bacterial cell envelope and associated extracellular structures serve as the primary interface between a bacterium and its environment, providing structural integrity and protection against osmotic pressure (StatPearls, NBK554425). This complex assembly typically includes the inner cytoplasmic membrane, a peptidoglycan cell wall, and, in Gram-negative bacteria, an outer membrane containing lipopolysaccharides (Nature Reviews Microbiology, nrmicro3238). Extracellular structures such as capsules, pili, and biofilm matrices further contribute to virulence, adhesion, and evasion of the host immune system (Microbiology Spectrum, 2017). Because many of these components are unique to prokaryotes, they represent critical targets for a wide range of antibiotics (Merck Manual, 2023). Drugs like beta-lactams and glycopeptides inhibit the synthesis of the cell wall, while polymyxins and lipopeptides disrupt the integrity of the cell membranes, leading to rapid bacterial cell death (PubMed, 26248330).
