Overview

The bacterial cell envelope, which includes the cell wall and the cytoplasmic (and in Gram-negative bacteria, the outer) membrane, is essential for maintaining bacterial structural integrity and regulating the internal environment [1]. The cell wall is primarily composed of peptidoglycan, a polymer of sugars and amino acids that provides mechanical strength and protects the cell from osmotic lysis [1][2]. The cell membrane serves as a selective barrier and a scaffold for essential processes like energy production and lipid synthesis [1]. Antimicrobial agents targeting these components are among the most successful clinical drugs; beta-lactams and glycopeptides inhibit cell wall synthesis, while polymyxins and lipopeptides disrupt membrane stability [2][3]. Because peptidoglycan is absent in human cells, cell wall inhibitors often exhibit high therapeutic indices, though membrane-disrupting agents can sometimes cause host toxicity due to similarities in lipid structures [2]. The continuous evolution of resistance mechanisms, such as modified penicillin-binding proteins or altered membrane charges, remains a primary challenge in targeting these structures [3].
