The bacterial cell membrane and intracellular components encompass a diverse array of structural and functional elements essential for bacterial viability and replication. The cytoplasmic membrane acts as a selective permeability barrier and a scaffold for bioenergetic processes, while intracellular components include the ribosomes, nucleic acids, and metabolic enzymes required for life. Antibiotics targeting the membrane, such as polymyxins and daptomycin, disrupt lipid organization or create pores, leading to the loss of essential ions and cell death (Zavascki et al., 2007; DOI: 10.1093/jac/dkl487). Intracellular targets are addressed by drugs that must cross the cell envelope to inhibit critical processes like protein synthesis at the ribosome or DNA replication via topoisomerase inhibition (Wilson, 2014; DOI: 10.1128/MMBR.00013-14). These targets are central to the treatment of a wide range of bacterial infections, from localized skin infections to systemic sepsis. However, the complexity of these systems and the rapid evolution of resistance mechanisms present significant therapeutic challenges. Effective drug design must balance potent antibacterial activity with minimal toxicity to host cells, which lack many of these specific bacterial structures (Bush et al., 2015; DOI: 10.1128/MMBR.00009-15).
