Overview

Bacterial cell structures and biofilms represent a broad category of therapeutic targets essential for bacterial survival and pathogenesis. The bacterial cell envelope, comprising the cell wall (peptidoglycan) and membranes, provides structural integrity and serves as a primary barrier against environmental stress; it is the target of major antibiotic classes like beta-lactams and glycopeptides (StatPearls, 2023). Biofilms are complex, multicellular communities of bacteria embedded within a self-produced matrix of extracellular polymeric substances (EPS), including polysaccharides, proteins, and extracellular DNA (Nature Reviews Microbiology, 2016). This biofilm architecture protects bacteria from host immune responses and increases antibiotic tolerance by up to 1,000-fold compared to planktonic cells (Journal of Bacteriology, 2019). Targeting these structures is critical for treating chronic and device-related infections, where traditional antibiotics often fail due to the protective nature of the biofilm matrix and the presence of persister cells.
