Bacterial cell wall peptidoglycan synthesis refers to the multi-step enzymatic process by which bacteria construct their essential cell wall component, peptidoglycan. Peptidoglycan is a mesh-like polymer composed primarily of alternating N-acetylglucosamine and N-acetylmuramic acid sugars cross-linked by short peptides. This structure provides mechanical strength, maintains cellular shape, and protects bacteria from osmotic lysis. The biosynthetic process involves cytoplasmic formation of nucleotide sugar precursors, membrane-associated assembly into lipid-linked intermediates, and extracellular polymerization with peptide cross-linking catalyzed by transpeptidases known as penicillin-binding proteins. Many clinically important antibiotics—including β-lactams and glycopeptides—target various stages in this pathway because it is essential for bacterial survival but absent in humans. Bacteria can develop resistance through modifications in enzymes or alternative cross-linking mechanisms.
