Overview

Bacterial cellular components and keratin proteins refers to the collective binding sites for triphenylmethane dyes, most notably gentian violet (also known as methylrosaniline chloride). This target profile is characterized by a high affinity for the negatively charged components of bacterial cells, such as the peptidoglycan layer in Gram-positive bacteria, as well as the keratin proteins found in human skin, hair, and nails (DrugBank, DB00406). By binding to these bacterial structures, the associated drugs exert potent antibacterial, antifungal, and anthelmintic effects, likely by interfering with cellular respiration or cell wall integrity (StatPearls, NBK553144). The simultaneous binding to host keratin allows the drug to remain localized at the site of infection, providing a sustained antiseptic effect, though it also causes the characteristic deep purple staining of the skin (PubChem, CID 11057). This target is not a single molecular entity but a description of the broad chemical affinity that mediates the therapeutic and physical properties of specific topical agents. Consequently, it is often used in the context of older antiseptic treatments for skin and mucous membrane infections, such as candidiasis or impetigo.
