Bacterial enzymes and membrane lipids refers to a broad collection of molecular structures and catalytic proteins that are essential for the survival, growth, and structural integrity of bacteria. Bacterial enzymes include critical proteins like penicillin-binding proteins (PBPs), which catalyze cell wall synthesis, and DNA gyrase, which is necessary for DNA replication and transcription (StatPearls, 2023). Membrane lipids, such as phospholipids and lipopolysaccharides (LPS), form the essential barrier of the bacterial cell envelope, protecting the organism from environmental stressors and host immune defenses (Nature Reviews Microbiology, 2017). Antibiotics target these components through various mechanisms: beta-lactams and glycopeptides inhibit cell wall enzymes, while lipopeptides and polymyxins directly interact with and disrupt the lipid bilayer (NIH, 2022). Although these are valid therapeutic targets, the term is considered too non-specific for precise drug-target mapping as it encompasses thousands of distinct molecular entities across different bacterial species. The clinical utility of targeting these structures is often limited by the emergence of antimicrobial resistance and the potential for off-target effects on the human commensal microbiome (WHO, 2021).
