Overview

Exotoxins are potent protein molecules secreted by various pathogenic bacteria, including both Gram-positive and Gram-negative species, to facilitate infection and host colonization. Unlike endotoxins, which are structural components of the bacterial cell wall, exotoxins are actively released into the surrounding environment and can cause extensive damage at sites distant from the infection (StatPearls, 2023). They are broadly categorized based on their mechanism of action, such as AB toxins that inhibit protein synthesis (e.g., Diphtheria toxin), pore-forming toxins that induce cell lysis (e.g., Alpha-hemolysin), and superantigens that cause massive immune overactivation (e.g., Toxic shock syndrome toxin-1) (NCBI, 2022). In clinical medicine, exotoxins are critical therapeutic targets; treatment often involves the administration of monoclonal antibodies or antitoxins that neutralize the toxin before it can bind to or enter host cells (PubMed, 2021). Additionally, chemically inactivated exotoxins, known as toxoids, serve as the primary components of highly effective vaccines for diseases like tetanus and diphtheria.
