Bacterial Membrane Lipid II (Lipid II)

Lipid II is a crucial precursor molecule in the biosynthesis of bacterial cell walls. It is an amphipathic peptidoglycan intermediate, characterized by a bactoprenol (undecaprenyl) hydrocarbon chain that serves as a lipid anchor, embedding the molecule within the bacterial cytoplasmic membrane. Lipid II plays an essential role in delivering and incorporating disaccharide-pentapeptide building blocks into the growing peptidoglycan mesh, which forms the rigid structure of bacterial cell walls. Its unique structure—absent from eukaryotes but essential for most bacteria—makes it an attractive target for antibiotics. Several classes of antibiotics bind directly to or disrupt processes involving lipid II.
