Overview

Bacterial enzymes and metabolic pathways represent a broad category of therapeutic targets essential for the survival and proliferation of pathogenic bacteria. These targets are exploited by various classes of antibiotics to achieve selective toxicity, as many bacterial enzymes and pathways differ significantly from those in human hosts (Study.com; Creative Biolabs). Key examples include the synthesis of the peptidoglycan cell wall, targeted by beta-lactams; the replication of DNA, targeted by fluoroquinolones; and the biosynthesis of folic acid, targeted by sulfonamides and trimethoprim (NIH; Pressbooks). These pathways are critical for maintaining cellular integrity, genetic fidelity, and metabolic homeostasis (NIH). However, the effectiveness of drugs hitting these targets is frequently compromised by the development of resistance, such as the production of inactivating enzymes like beta-lactamases or modifications to the target enzymes themselves (NIH; News-Medical). Monitoring efficacy often involves measuring the minimum inhibitory concentration (MIC) or tracking systemic inflammatory markers like C-reactive protein (NIH). Despite their success, therapeutic challenges include the disruption of the host's commensal microbiome and the continuous evolution of multidrug-resistant strains (NIH; Iberoamerican Journal of Medicine).
