Overview

The bacterial respiratory chain in Staphylococcus spp. is a vital metabolic system responsible for generating ATP and maintaining the cell's redox state. In Staphylococcus aureus, this pathway is distinct because it utilizes menaquinone as the primary electron carrier, making it a vulnerable target for specific inhibitors. The "red benzoquinone" (3,5-dimethoxy-2-(methylthio)cyclohexa-2,5-diene-1,4-dione), a compound isolated from the venom of the scorpion Diplocentrus melici, has been identified as a potent antimicrobial agent targeting this system. It acts by disrupting electron flow and inducing the production of reactive oxygen species (ROS), which cause catastrophic oxidative damage to the bacterial cell. This mechanism is effective even against multidrug-resistant (MDR) strains of Staphylococcus, highlighting its potential as a lead for new antibiotic development. However, therapeutic application is challenged by the need for high selectivity to avoid toxicity to human cells, as some benzoquinone derivatives have shown cytotoxic effects on human mononuclear cells in vitro.
