Overview

The **bacterial ribosomal 30S subunit** is the smaller component of the prokaryotic 70S ribosome. It consists primarily of a single molecule of **16S rRNA** and approximately **21 proteins**. The main function of this macromolecular complex is to bind messenger RNA (mRNA) and facilitate its decoding during translation, ensuring accurate initiation at the start codon via recognition of sequences such as Shine-Dalgarno. The 30S subunit plays an essential role in assembling with the larger 50S subunit to form an active ribosome for protein synthesis. Many clinically important antibiotics—including tetracyclines and aminoglycosides—exert their antibacterial effects by binding specifically to sites on this subunit, thereby inhibiting bacterial growth through disruption of translation[1][2][3][4].

Mechanism of action

Inhibition of protein synthesis by blocking tRNA binding or causing misreading of mRNA[4] - Tetracyclines block the attachment of aminoacyl-tRNA to the A site on the ribosome. - Aminoglycosides interfere with proofreading, leading to errors in protein synthesis.
