Overview

Small molecule 0390 (6056-0390) is a novel triazoloquinazoline derivative identified through high-throughput screening for its potent and specific antimicrobial activity against Escherichia coli (Li et al., 2022) [11]. Although its exact molecular targets have not yet been definitively identified, experimental data suggests that the compound acts on essential intracellular processes, with current hypotheses focusing on the inhibition of protein synthesis, DNA replication, or the induction of reactive oxygen species (ROS) (Li et al., 2022) [1, 11]. The compound's efficacy is significantly enhanced when used in combination with the outer membrane-disrupting agent SPR741, suggesting that 0390 requires access to the bacterial cytoplasm to exert its bacteriostatic effects (Li et al., 2022) [1, 23]. Notably, 0390 demonstrates a high degree of species specificity, showing little to no activity against other common pathogens like Pseudomonas aeruginosa or Staphylococcus aureus (Li et al., 2022) [11]. Preliminary in vitro and in vivo studies indicate that 0390 possesses a favorable safety profile, with minimal cytotoxicity to human cells and no significant hemolytic activity (Li et al., 2022) [11, 37]. These findings position the uncharacterized targets of 0390 as viable candidates for the development of next-generation antibiotics to combat multidrug-resistant E. coli infections (Li et al., 2022) [11].
