Bacterial virulence and toxin production pathways represent a diverse set of therapeutic targets aimed at disarming pathogens rather than killing them directly (PMC5419286). This anti-virulence strategy focuses on neutralizing factors that facilitate host colonization, tissue damage, and immune evasion, such as exotoxins, quorum-sensing (QS) systems, and specialized secretion systems like the Type III Secretion System (T3SS) (PMC8538941). By targeting these non-essential pathways for bacterial growth, these therapies exert less selective pressure for the development of antimicrobial resistance compared to traditional bactericidal or bacteriostatic antibiotics (Nature Reviews Microbiology, 2025). Key examples include monoclonal antibodies that neutralize specific toxins, such as bezlotoxumab for Clostridioides difficile, and small molecules that disrupt bacterial communication or adhesion (Frontiers in Microbiology, 2021). While promising, these targets often require pathogen-specific diagnostics and early intervention to be effective in clinical settings (GARDP Revive, 2024).
