Bactoprenol is a C55 isoprenoid alcohol that serves as a vital lipid carrier in the biosynthesis of the bacterial cell wall (PubChem CID 5460541). It functions by transporting hydrophilic precursors, such as peptidoglycan and teichoic acid subunits, across the cytoplasmic membrane to the external assembly site (Stone & Strominger, 1971). The molecule cycles between various phosphorylated states, with undecaprenyl phosphate being the primary functional form that binds to cell wall building blocks. This transport mechanism is essential for maintaining the structural integrity of both Gram-positive and Gram-negative bacteria. Because bactoprenol-mediated transport is unique to prokaryotes, it represents a highly selective target for antimicrobial therapy. Antibiotics like bacitracin inhibit the recycling of this carrier by preventing the dephosphorylation of undecaprenyl pyrophosphate (Stone & Strominger, 1971). Other agents, such as friulimicin B, directly sequester the lipid carrier to halt cell wall production (Schneider et al., 2009). The disruption of this cycle leads to the accumulation of precursors and eventual bacterial cell lysis.
