Baculoviral IAP repeat-containing protein 8 (BIRC8)

Target
BIRC8
Molecular classification
Other (IAP family protein; not a receptor, enzyme, transporter, or classical drug target)
01

Overview

Baculoviral IAP repeat-containing protein 8 (BIRC8) is a member of the inhibitor of apoptosis protein (IAP) family, containing a baculoviral IAP repeat (BIR) domain and predicted E3 ubiquitin ligase activity. It is thought to protect against apoptosis mediated by BAX and promote protein ubiquitination, and is associated with negative regulation of apoptosis. However, functional and disease relevance in humans remain unclear—BIRC8 is listed as a pseudogene in some sources and does not appear to be a validated therapeutic target. The BIRC8 gene product is mainly found in the cytoplasm and nucleus and shares homology with other IAPs, which are well-known regulators of cell death[1][4][5][3].

02

Mechanism of action

Not established for BIRC8; general IAP antagonism is via disruption of caspase inhibition, but this has not been described for BIRC8 as a drug target

03

Biological functions

Negative regulation of apoptotic processUbiquitin ligase activityPositive regulation of Wnt signalingPositive regulation of protein ubiquitination
04

Disease associations

Cancer (noted in related literature for IAP family, but unclear for BIRC8 specifically)
05

Safety considerations

None known; no drugs in clinical trials target BIRC8 directly, so no safety profile is established
06

Interacting drugs

None known for BIRC8 specifically; general IAP inhibitors (such as SMAC mimetics) do not directly target BIRC8 (see notes below)
07

Biomarkers

None known for BIRC8 specifically
