BAI1-associated protein 3 (BAIAP3) is a C2 domain-containing protein highly expressed in brain, encoded by the BAIAP3 gene on human chromosome 16[1][2][3][7]. It was identified as an interacting partner of brain-specific angiogenesis inhibitor 1 (BAI1), a p53-inducible protein. BAIAP3 is involved in intracellular membrane trafficking, specifically mediating endosome to Golgi retrograde transport. This process is calcium-dependent and may involve interactions with SNARE fusion receptors and membrane phospholipids[3][7]. By promoting the recycling of secretory vesicle membrane proteins, BAIAP3 indirectly regulates the maturation and exocytosis of dense-core vesicles that release neurotransmitters and hormones, such as neuropeptide Y (NPY), serotonin, and insulin[3][7]. BAIAP3's functions are critical for synaptic transmission and possibly for regulating behaviors linked to neurotransmitter secretion[4][5]. While BAIAP3 is associated with certain diseases, including neuropsychiatric disorders and hereditary mixed polyposis syndrome, there is little evidence it is currently a direct therapeutic target or is drugged[3].
