Overview

BarH like homeobox 1 (BARHL1) is a DNA-binding transcription factor from the homeobox family, critical for CNS development, sensory cell differentiation, and neural fate specification. The protein is highly expressed in neural precursors, the developing inner ear, and migration zones of the embryonic brain, supporting neuronal progenitor generation and the long-term maintenance of auditory hair cells vital for hearing and balance. Mutations or altered expression of BARHL1 are linked to neurodevelopmental disorders such as Joubert syndrome, and its expression is associated with cancer progression and neurodegenerative diseases. While a potential research and diagnostic target, it is not currently modulated by any approved drugs, and direct therapeutic targeting could entail CNS-specific safety challenges.
