BARX homeobox 2 (BARX2) is a transcription factor belonging to the Bar-like homeobox gene family, characterized by a homeobox DNA-binding domain. It binds to specific DNA sequences to regulate gene expression involved in diverse biological processes such as cell proliferation, differentiation, immune response, and tumorigenesis[1][2][6]. BARX2 plays a significant role in embryonic development, regulation of cell adhesion molecules (notably in neural and epithelial tissues), and actin cytoskeleton remodeling[2][6]. It is implicated in the regulation of keratin and neural adhesion molecule genes, and is crucial for mesenchymal aggregation and cartilage differentiation[5]. Dysregulation of BARX2 has been linked to various cancers (including ovarian, gastric, lung, and liver cancers), and diseases such as Jacobsen Syndrome and craniosynostosis[1][2]. High BARX2 expression can serve as a biomarker for poor prognosis in certain cancers such as hepatocellular carcinoma[1]. Currently, no approved drugs or direct pharmacological modulators of BARX2 are documented, but its role in cancer pathways suggests potential as a future therapeutic target[1][2][6].
