Basal body orientation factor 1 (BBOF1) is a structural protein involved in cilia function and sperm motility. It stabilizes the axoneme—the core structure of cilia and flagella—thus supporting proper alignment and movement in multiciliated cells, as well as sperm motility and male fertility. The protein features two coiled-coil domains and a domain of unknown function (DUF4515/pfam14988)[1]. BBOF1 is predicted to stabilize key proteins (ODF2, MANS1) associated with sperm flagella[2][7]. It is expressed in testis, cerebellum, and lung tissues, and interaction data suggests links to DNA Ligase IV, NRF1, and HYLS1[1]. No clinical drugs or therapeutic actions are currently associated with BBOF1, and it is not recognized as a standard therapeutic target. While linked to sperm function and possibly some disease contexts (such as glioma or metabolic deficiency), it primarily acts as a structural and regulatory protein in ciliated cells[1][2][7].
