Basic salivary proline-rich protein 1 (PRB1) is a major component of salivary proline-rich proteins (PRPs) in humans. Encoded by the PRB1 gene, it belongs to a family of polymorphic, secreted, intrinsically unstructured proteins characterized by high proline content and repetitive sequence motifs. PRB1 is involved in the maintenance of oral health, contributing to the formation of the enamel pellicle, lubricating oral surfaces, binding plant polyphenols (tannins), and modulating microbial adhesion, thereby playing a protective role in the oral cavity. The PRB1 gene is clustered with other PRP genes on chromosome 12p13, which also encodes related proteins with similar structural and functional properties. PRB1 is not known to be a classical therapeutic target such as a receptor, transporter, or enzyme, and no drugs currently target it directly. However, PRB1 genetic variants have been studied for their polymorphisms and potential associations with oral or systemic diseases.
