Basonuclin zinc finger protein 2 (BNC2) is an evolutionarily conserved zinc finger transcription factor essential for vertebrate development, especially craniofacial morphogenesis, pigmentation, and tissue homeostasis[1][4][6]. It consists of three pairs of zinc finger motifs, a serine-rich region, and a nuclear localization signal, and is primarily localized to the nucleus[3][6]. BNC2 is widely expressed in various tissues, including reproductive organs, kidney, intestine, uterus, and skin, and appears to regulate the rate of cell multiplication in mesenchymal cells during embryogenesis[1][6]. Human mutations or regulatory variants in BNC2 have been linked with craniofacial disorders (such as cleft palate), pigmentation traits, and susceptibility to adolescent idiopathic scoliosis[4][5]. BNC2 does not currently serve as a direct therapeutic target, and there are no drugs known to act selectively on this transcription factor. Its broad developmental roles and association with disease phenotypes underscore its functional importance in human biology and disease[1][4][5][6].
