Overview

Bassoon presynaptic cytomatrix protein (BSN) is a large scaffolding protein located at the presynaptic active zone of neurons where neurotransmitter release occurs[1][6]. It organizes the cytomatrix underlying the plasma membrane and ensures proper spatial arrangement of synaptic vesicle clusters and other protein complexes critical for synaptic vesicle fusion and endocytosis[1][2][6]. BSN is primarily expressed in the brain, is highly concentrated at active zones, and is essential for regulating neurotransmitter release. It interacts with other cytomatrix proteins and contributes to synaptic targeting, assembly of the active zone, and communication from synapse to nucleus, modulating gene expression and presynaptic autophagy. Mutations or dysfunctions in this protein have been associated with epilepsy and autism spectrum disorder[1]. No drugs are known to directly target this protein, nor is it considered a therapeutic target in current pharmacology, but it plays a crucial structural and regulatory role in neuronal function[1][6].
