BCAN antisense RNA 1 (BCAN-AS1)

Target
BCAN-AS1
Molecular classification
Long non-coding RNA (lncRNA), Antisense RNA, Non-coding RNA
01

Overview

BCAN antisense RNA 1 (BCAN-AS1) is a long non-coding antisense transcript implicated in oncogenesis, particularly in pancreatic ductal adenocarcinoma. Mechanistically, BCAN-AS1 acts as a scaffold that stabilizes the c-Myc protein by facilitating its interaction with SNIP1 and preventing SKP2-mediated c-Myc ubiquitination and degradation. BCAN-AS1 overexpression is associated with increased tumor aggressiveness and poor prognosis. Knockdown of BCAN-AS1 using antisense oligonucleotides suppresses tumor growth and metastasis in model systems, suggesting therapeutic potential for cancers with high BCAN-AS1 expression.

02

Mechanism of action

Antisense oligonucleotide inhibition leads to reduced BCAN-AS1 RNA levels, thereby destabilizing c-Myc protein and inhibiting tumor growth and metastasis

03

Biological functions

Regulation of gene expression (notably c-Myc stabilization)Promoting malignant phenotypesModulation of protein ubiquitination and degradation (by blocking SKP2-mediated c-Myc ubiquitination)Acting as a molecular scaffold
04

Disease associations

Cancer, specifically pancreatic ductal adenocarcinoma (PDAC)Associated with poor prognosis in cancerAlso linked to other cancers such as lung cancer and cervical cancer (per associative databases)
05

Safety considerations

No direct safety concerns reported; general challenges apply to targeting lncRNAs, including delivery and potential off-target effects
06
