Overview

BCDIN3 domain containing RNA methyltransferase (BCDIN3D) is an enzyme of the Rossmann fold methyltransferase family that functions mainly as a 5'-methylphosphate capping enzyme for cytoplasmic histidyl tRNA (tRNA(His))[1][2][5]. By using S-adenosylmethionine as a methyl donor, it monomethylates the 5'-monophosphate of tRNA(His), a modification that protects the tRNA from degradation. BCDIN3D can also methylate the 5'-monophosphate of some precursor microRNAs (pre-miRNAs), acting as a negative regulator of miRNA processing, by interfering with Dicer-mediated cleavage and maturation—this mechanism appears particularly important in cancer cells, such as breast cancer, in which BCDIN3D is overexpressed and is associated with tumor invasiveness and poor prognosis. It may thus play roles in tumorigenesis through post-transcriptional regulation of gene expression[1][2][3][5].
