Overview

BCL2L1 pre-mRNA is the unspliced RNA transcript produced from the BCL2L1 gene. Alternative splicing of this transcript generates two key protein isoforms: Bcl-xL, which prevents cell death and is overexpressed in many cancers, and Bcl-xS, which promotes apoptosis. Modulating the splicing of BCL2L1 pre-mRNA represents a therapeutic strategy to shift the balance toward the pro-apoptotic isoform, thereby restoring apoptosis in cancer cells that evade cell death. Pre-mRNA-based targeting is distinct from classical drug targeting of the mature protein, focusing on the regulatory RNA stage to effect isoform switching[5][7]. BCL2L1 pre-mRNA itself is not a traditional drug target (such as a receptor or enzyme), but it is an emerging therapeutic target through modulation of RNA splicing. Most drugs in clinical use target the mature protein (Bcl-xL), while investigational therapies attempt to modify splicing at the pre-mRNA level, especially in cancer[7]. The term "BCL2L1 pre-mRNA" may be considered imprecise for protein-centric target lists, as the disease-modifying activity is ultimately due to modulation of protein isoform production[7][5].
