Overview

BCL2 interacting protein 1 (BNIP1) is a mammalian ortholog of yeast Sec20p and a member of the BH3-only protein family, primarily defined by its capability to interact with antiapoptotic proteins such as BCL2 and adenovirus E1B 19-kDa protein[2][1]. BNIP1 is an integral component of the syntaxin 18 complex, which mediates ER membrane fusion and organization. It plays pivotal roles in apoptosis—mediated by its BH3 domain—and in maintaining the ER network, likely by facilitating membrane fusion through interactions with α-SNAP and other SNARE proteins[1][2][4]. While mutations of BNIP1 may be associated with certain genetic diseases, it is also notable as a proapoptotic molecule, suggesting relevance in cancer biology and potential as a therapeutic target, although no drugs directly targeting BNIP1 are currently established.
