Overview

BCL6 corepressor (BCOR) is a ubiquitously expressed protein that acts as a transcriptional corepressor by interacting directly and selectively with the POZ domain of the BCL6 transcription factor[1][2]. BCOR regulates gene expression by recruiting histone deacetylases, thereby modifying chromatin to suppress target genes. It is essential for embryogenesis, including development of eyes, heart, teeth, and other organs, and appears to participate in apoptotic regulation[1][3][4]. Pathogenic mutations in the BCOR gene cause a spectrum of severe congenital syndromes, particularly affecting females, and are also implicated in hematologic malignancies such as acute myeloid leukemia[2][3][4]. Interactions of BCOR are highly specific; it binds BCL6 and selected epigenetic regulators (HDACs) but not most other POZ proteins[1].
