BDP1 general transcription factor IIIB subunit (BDP1) is a transcription factor that is a core component of the TFIIIB complex, which also includes Brf1/Brf2 and TBP (TATA-binding protein)[1][2][4][5]. TFIIIB is essential for the recruitment of RNA polymerase III (Pol III) to its target genes and is critical for the transition from a closed to an open pre-initiation complex during transcription initiation. BDP1 plays a central, unique role in this process that is not substituted by other factors in the Pol I or Pol II transcription systems. Structurally, BDP1 contains a highly conserved SANT domain responsible for interactions with Brf1/Brf2-TBP complexes and the promoter DNA. It stabilizes the TFIIIB-DNA complex and is required for DNA strand separation, a key step in initiating Pol III transcription. Functional studies highlight its essential role in Pol III transcription and its involvement in the cellular response to proliferative signals and tumorigenesis, such as promoting cancer progression in P53-mutated prostate cancers[1][3][4][5]. Notes: - BDP1 is not a classical drug target (i.e., not a receptor or enzyme typically targeted by small molecules or biologics); no interacting drugs or specific mechanisms of pharmacological action have been reported in the literature as of now. - It is sometimes referred to as "TFIIIB150" reflecting its molecular weight. - It is not to be confused with the similarly named BDP1 gene/protein found in other transcriptional or chromatin-related complexes; all listed aliases are for this particular human RNA polymerase III-associated factor.
