The Beclin-1·B-cell lymphoma 2 (Bcl-2) complex is a pivotal regulatory assembly that governs the crosstalk between autophagy and apoptosis (Pattingre et al., 2005, Cell). Beclin-1 serves as a core component of the Class III Phosphoinositide 3-kinase (PI3K) complex required for autophagosome formation, while Bcl-2 is a well-characterized anti-apoptotic protein (Sinha & Levine, 2008, Oncogene). When Bcl-2 binds to the BH3 domain of Beclin-1, it sequesters the protein and inhibits its ability to initiate autophagy, thereby acting as a molecular switch between cell survival and programmed cell death (Oberstein et al., 2007, J. Biol. Chem.). This interaction is crucial for maintaining cellular homeostasis under various stress conditions, including nutrient deprivation and hypoxia (He et al., 2012, Nature). In many cancers, the overexpression of Bcl-2 leads to the pathological suppression of autophagy, contributing to tumor progression and resistance to chemotherapy (Kang et al., 2011, Autophagy). Conversely, in neurodegenerative diseases, enhancing the dissociation of this complex can promote the clearance of toxic protein aggregates through the autophagic pathway (Salminen et al., 2013, Progress in Neurobiology). Pharmacological targeting of this complex typically involves BH3 mimetics or specific peptides designed to disrupt the protein-protein interaction, thereby restoring autophagic flux or inducing apoptosis in malignant cells (Shoji-Kawata et al., 2013, Nature).
