The Beclin 1–Vps34 complex is a core multi-protein assembly essential for the initiation of autophagy and endocytic trafficking in mammalian cells. It consists primarily of Beclin 1 (BECN1), a scaffold protein, and Vps34 (phosphatidylinositol 3-kinase catalytic subunit type 3, PIK3C3), a lipid kinase. The complex also includes regulatory subunits such as Vps15 (PIK3R4), ATG14 or UVRAG, and sometimes accessory proteins like NRBF2. The Beclin 1–Vps34 complex catalyzes the generation of PtdIns(3)P, a lipid signal crucial for recruiting proteins that mediate autophagosome biogenesis and membrane trafficking. Distinct combinatorial assemblies (Beclin 1–Vps34–ATG14 and Beclin 1–Vps34–UVRAG) direct the complex toward autophagy versus endocytosis, respectively. Dysregulation of this complex is implicated in cancer, neurodegenerative diseases, and infections. The complex is a validated therapeutic target for modulating autophagy, though direct pharmacological targeting is still experimental. Inhibitors of VPS34 kinase activity are being developed as modulators of autophagy in disease contexts[1][2][5][4][6].
