Overview

Beclin 1 (BECN1) mRNA is the transcript encoding the Beclin 1 protein, a central regulator of macroautophagy and a key member of the Class III phosphatidylinositol 3-kinase (PI3K) complex. This protein acts as a scaffold, coordinating the assembly of various co-factors that initiate autophagosome formation, a process vital for maintaining cellular homeostasis by degrading damaged organelles and misfolded proteins (UniProt Q14457). In clinical contexts, BECN1 is recognized as a potent tumor suppressor; its expression is frequently reduced in breast, ovarian, and prostate cancers, where its loss correlates with increased genomic instability and tumor progression (PubMed: 10619014). In neurodegenerative conditions, such as Alzheimer's disease, BECN1 levels are often diminished, leading to the toxic accumulation of amyloid-beta and tau proteins (PubMed: 16763144). Therapeutic strategies targeting BECN1 mRNA involve the use of RNA interference (RNAi) or antisense oligonucleotides (ASOs) to either silence its expression in specific pathological contexts or, more commonly in research, to investigate the consequences of autophagy inhibition.
