Overview

BEN domain-containing protein 4 (BEND4) is a DNA-binding protein predicted to participate in chromatin organization and transcriptional regulation[2][3][5]. It is a member of the BEN domain family, which is involved in chromatin remodeling and epigenetic control of gene expression[1]. BEND4 is expressed in a variety of human tissues, including the testis and several cancer types[7]. Elevated BEND4 expression is implicated in oncogenic processes, particularly as an independent negative prognostic marker in diffuse large B-cell lymphoma (DLBCL), where it is associated with poor survival, suppressed immune infiltration, and chemoresistance[1]. While not currently a direct target of approved drugs, BEND4 modulation affects cellular responses to chemotherapy[1]. There is also preliminary evidence for BEND4 involvement in other diseases, including retinitis pigmentosa and infection-related encephalopathy, though these roles require further characterization[2][4].

Mechanism of action

Not a direct drug target in standard therapies; but BEND4 knockdown increases sensitivity to certain chemotherapeutic agents (e.g., doxorubicin) in lymphoma models[1]. No direct-acting small molecule or biologic drugs reported.
