Overview

BEN domain-containing protein 5 (BEND5) is a sequence-specific transcriptional repressor that possesses a BEN domain, enabling direct DNA binding to defined motifs. BEND5 serves as a chromatin boundary factor and negatively regulates DNA-templated transcription. It is particularly implicated in neurogenesis and early germ cell differentiation, functioning by demarcating chromatin boundaries and promoting the transition of pluripotent cells toward specific developmental fates. Aberrant expression of BEND5 has been associated with pathologic tissue states (such as arachnoid cysts) and chromosomal rearrangements, including a role as a fusion partner in BEND5–ALK fusions found in pediatric cancers, suggesting a possible but as yet unexploited disease association. BEND5 is not currently considered a direct therapeutic target, and no drugs are known to interact with it[1][2][3][4][5].
