Overview

BEN domain-containing protein 6 (BEND6) is a nuclear co-repressor that binds the CSL transcription factor (CBF1/RBPJ) and antagonizes Notch-dependent gene activation, thereby inhibiting Notch-mediated self-renewal and promoting neural differentiation. It acts analogously to the *Drosophila* Insensitive protein as a BEN-solo factor regulating neural cell fate, and is thought to fine-tune Notch signaling during embryonic brain development. BEND6 is predicted to act within the nucleus, regulating chromatin and gene expression relevant to neural stem cell differentiation. Diseases associated include Carpenter syndrome 1, and while it modulates pathways central to neurodevelopment, it is not currently known to be a direct target of approved drugs[1][2][3][4][5].
