Overview

Beneficial bacteria, commonly known as probiotics or commensal microbiota, are live microorganisms that provide health benefits to the host by maintaining physiological homeostasis and supporting the 'microbial organ' (Source 1.1.1, 1.4.4). They are primarily located in the gastrointestinal tract, where they assist in nutrient metabolism, synthesize essential vitamins like B12 and K, and provide a robust defense against pathogenic colonization through competitive exclusion (Source 1.1.2, 1.1.3). In the context of disease, a disruption in the balance of these bacteria, known as dysbiosis, is associated with a wide range of conditions including inflammatory bowel disease, metabolic disorders, and recurrent infections like Clostridioides difficile (Source 1.2.1, 1.4.1). Therapeutic strategies increasingly focus on modulating these populations through Live Biotherapeutic Products (LBPs) or fecal microbiota transplants to restore microbial diversity and function (Source 1.2.2, 1.2.5). Mechanism of action

Beneficial bacteria exert therapeutic effects through competitive exclusion of pathogens for nutrients and adhesion sites, production of short-chain fatty acids (SCFAs) like butyrate that serve as energy for colonocytes, and modulation of the host immune system by inducing regulatory T cells and anti-inflammatory cytokines like IL-10 (Source 1.1.2, 1.3.5). They also enhance the intestinal epithelial barrier by upregulating tight junction proteins and produce antimicrobial peptides known as bacteriocins to directly inhibit harmful microbes (Source 1.1.2, 1.3.4).
