The benzylpenicilloyl-specific immunoglobulin E antibody is an IgE-class antibody produced by the immune system in some individuals after exposure to benzylpenicillin or related β-lactam antibiotics. It specifically recognizes the benzylpenicilloyl determinant, a major antigenic epitope formed when penicillin reacts with proteins in the body. This antibody is responsible for mediating the immediate hypersensitivity reactions (Type I allergic responses) seen in penicillin allergy, including urticaria, angioedema, bronchospasm, and potentially life-threatening anaphylaxis. Diagnostic skin tests use benzylpenicilloyl polylysine to detect its presence. Laboratory assays (such as RAST) can quantify BPO-specific IgE in patient serum as a biomarker of penicillin allergy. The presence of this antibody is not considered a drug target for therapeutic intervention but is crucial for allergy diagnosis and patient safety.
