The BET (Bromodomain and Extra-Terminal domain) family of proteins is a group of epigenetic "reader" proteins that play a central role in regulating gene transcription through their interactions with acetylated lysine residues on histones. The mammalian BET family consists of four members: BRD2, BRD3, BRD4 (ubiquitously expressed), and BRDT (testis-specific expression). They function in reading histone acetylation marks, recruiting key transcription factors, and remodeling chromatin structure. They are involved in diverse cellular processes such as cell cycle progression, cellular proliferation and differentiation, inflammation response, and neurogenesis, and play roles in diseases such as cancer and viral latency.
BET proteins act as scaffolds for assembling the transcriptional machinery at active chromatin regions by reading histone acetylation marks, recruiting transcription factors, remodeling chromatin structure, and facilitating super-enhancer assembly.
