BET1L (Bet1 golgi vesicular membrane trafficking protein like) encodes a vesicular SNARE protein essential for proper membrane trafficking within the early secretory pathway, particularly in assembly and function of the Golgi SNARE complexes[1][2][4]. BET1L enables SNAP receptor activity and is involved in regulation of retrograde vesicle-mediated transport from the Golgi to the endoplasmic reticulum (ER) and endosome to Golgi trafficking. It localizes to the Golgi apparatus and endosomes[1][2]. BET1L has emerged as a genetic susceptibility locus for uterine fibroids and serves as a biomarker for prognosis in endometrial adenocarcinoma. Variations in its gene sequence, especially rs2280543, confer protection against specific fibroid subtypes. Altered BET1L levels have also been found in neurodegenerative disease models and glycosylation disorders, highlighting a role in cellular trafficking and disease pathophysiology[1][2][4]. There are no clinically established drugs targeting BET1L, nor established safety concerns directly related to its targeting. BET1L directly interacts with other vesicle fusion SNAREs such as GOSR1, STX5, and YKT6[4].
