Beta-1,4-mannan is a linear polysaccharide composed of D-mannose units linked by β-1,4-glycosidic bonds, serving as a major structural and storage component in plant cell walls and seeds, such as those of coffee, soy, and ivory nut (Wikipedia; NIH). In human health, Beta-1,4-mannan and its hydrolyzed derivatives, mannooligosaccharides (MOS), act as potent prebiotics that selectively promote the growth of beneficial gut bacteria like Bifidobacterium and Roseburia, leading to the production of health-promoting short-chain fatty acids (MDPI; NIH). Furthermore, Beta-1,4-mannan is an immunostimulatory molecule that binds to the TLR4/MD-2 complex and the complement C3a receptor (C3aR) on dendritic cells, inducing the secretion of pro-inflammatory cytokines such as IL-6 and TNF-α (NIH). This immunological activity has led to its exploration as a vaccine adjuvant and an immune-boosting functional food component (NIH). Additionally, the Beta-1,4-mannan backbone is a key component of galactomannan, a vital diagnostic biomarker for invasive aspergillosis (Wikipedia; ResearchGate). While not a traditional pharmacological target, it is the primary substrate for therapeutic and industrial enzymes like Beta-mannanase, which are used to improve digestion and facilitate targeted drug delivery (NIH; ResearchGate).
