Overview

Beta-1-syntrophin (SNTB1) is a peripheral membrane and cytoskeletal scaffold protein encoded by the SNTB1 gene. It is a member of the syntrophin family, which interacts with dystrophin, components of the dystrophin-associated glycoprotein complex, and various intracellular signaling and membrane proteins. SNTB1 contains two pleckstrin homology (PH) domains, a PDZ domain, and a syntrophin-specific region, supporting complex protein-protein interactions. It organizes the subcellular localization of key membrane proteins, linking them to the actin cytoskeleton. SNTB1 is primarily expressed in muscle, liver, kidney, the neuromuscular junction, and the retina. Loss of SNTB1 is associated with muscular dystrophies; recent research implicates SNTB1 in colorectal cancer, where its elevated expression promotes tumorigenesis by enhancing the Wnt/β-catenin signaling cascade, maintaining cancer stemness, and correlating with poor prognosis. SNTB1 represents a promising therapeutic target and biomarker in oncology, specifically colorectal cancer, though it is not yet a direct target of approved drugs and its physiological details in disease remain under investigation.
