Overview

Beta-2-syntrophin (SNTB2) is a 59 kDa peripheral membrane-associated adapter protein encoded by the SNTB2 gene and is a member of the syntrophin family, which includes several structurally related proteins. Syntrophin beta 2 is characterized by multiple protein-protein interaction domains, notably two pleckstrin homology (PH) domains, a PDZ domain, and a C-terminal syntrophin unique (SU) domain. It is predominantly expressed in skeletal and cardiac muscle, where it serves as a scaffold protein within the dystrophin-associated protein complex (DAPC), organizing membrane protein localization, linking receptors to the actin cytoskeleton, and assisting in the functional architecture of muscle cell membranes. SNTB2 is involved in processes such as regulation of signaling complexes and maintenance of membrane structural integrity. Mutations or dysfunction of the dystrophin-associated protein complex, including beta-2-syntrophin, are implicated in neuromuscular diseases like muscular dystrophy. Syntrophin beta 2 is not a direct drug target or therapeutic receptor, but its roles in protein trafficking, cytoskeletal organization, and membrane stability are crucial for cell function in muscle and possibly other tissues.
